Running a mile will use your aerobic energy system which will allow you to burn fat.
Doing any task which requires energy will burn calories (a calorie is effectively a measurement of energy), but a one mile run and a reasonable pace will burn more than a one mile race at a slow pace.

Running a mile burns 200 calories so yeah it burns calories.
Of course you burn calories! But if you want to lose pounds, do it atleast 3 times a week minimum for at least 20 minutes or more! :]
