I hate to follow the crowd, but yep, squats and lunges are the way to go!
Both work the glutes significantly.
Do you own a barbell? It might benefit you to be able to increase the resistance of each movement.

Pilates and jogging/running, bicycling, going up stairs, etc. It all uses your thighs and glutes.
Squats, lunges, stairs two or more at a time, walking on an incline, roller blading and backward leg lifts.
Everyone is spot on with squats, lunges, etc. If you go to a gym, use the butt machines. I don't know what it's called, but I call it the J Lo machine. It is by far the best. You lay on your stomach and lift the weights up to your butt with your legs. After a week I noticed a difference. Even my husband had a J Lo bootie after using it.
